This study aimed to characterize the metabolomic profile of aqueous extracts of Hypericum perforatum (St. John’s wort) commercial samples using 1H NMR spectroscopy, in order to evaluate their chemical composition as consumed in infusion form. Seven retail samples were extracted with water under conditions simulating tea preparation and analyzed using 1H NMR. Metabolite identification and quantification were performed with Chenomx® software, followed by multivariate statistical analyses including PCA and OPLS-DA. A total of 37 metabolites were identified, predominantly primary metabolites such as sugars (glucose, fructose), amino acids (glycine, asparagine), short-chain organic acids (malic, citric, and acetic acids), secondary metabolites (trigonelline, chlorogenic acid) and metabolism products (methanol, ethanol, acetic acid and acetone). Notably, key bioactive compounds traditionally associated with H. perforatum, such as hypericin and hyperforin, were not detected under the experimental conditions employed. Multivariate analyses revealed significant variability among samples, suggesting differences in composition likely related to origin, processing, or potential adulteration. These findings demonstrate that aqueous preparations of St. John’s wort differ substantially from extracts used in clinical trials, being dominated by primary metabolism rather than specialized bioactive compounds. This has important implications for the interpretation of its therapeutic effects when consumed as tea. Furthermore, the study highlights the utility of NMR-based metabolomics as a robust tool for quality assessment and authentication of herbal products

Background: Osteoporosis is a major health issue that has implications worldwide, both clinically and socioeconomically. Therefore, searching for informative bone health biomarkers that provide early information on bone changes and assess treatment efficacy is essential. Previously, our group used label-free proteomics and bioinformatic approaches to analyze the serum proteomic profile of postmenopausal women with normal bone mineral density (BMD), osteopenia, and osteoporosis. Based on their role in bone metabolism and literature review, a panel of 12 candidate serum biomarkers for low BMD was proposed.Objective: To validate a set of 6 proteins as potential minimally invasive serum biomarkers, Gelsolin (GSN), S*x Hormone-Binding Globulin (SHBG), Apolipoprotein A1 (APOA1), Apolipoprotein A2 (APOA2), Ryanodine Receptor 1 (RYR1), and Ryanodine Receptor 3 (RYR3), for diagnosing low BMD in postmenopausal women. We also investigated whether a combination of these proteins can provide more informative insights into the risk of having low BMD compared to individual biomarkers.Methods: The enzyme-linked immunosorbent assay was carried out to measure the concentrations of these proteins in 456 serum samples from postmenopausal Mexican women with normal or low BMD.Results: A random forest classifier demonstrated that SHBG and APOA1 are the most effective biomarkers for discriminating between women with normal or low BMD, with notable high sensitivity (86%), specificity (75%), precision (92%), and overall performance (89.32%).Conclusions: The combination of APOA1 and SHBG correlates better with the risk of bone loss than the performance of single biomarkers in postmenopausal women. Further validation in a larger cohort is necessary to confirm these results.

Anuran skin secretions are rich sources of bioactive metabolites whose composition may vary according to environmental conditions. In this study, skin secretions from two populations of Dendropsophus columbianus exposed to contrasting environmental conditions (Clarete and Las Guacas, Popayán, Cauca, Colombia) were evaluated for antifungal activity against Hemileia vastatrix, the causal agent of coffee leaf rust. Muicle (Justicia spicigera) is a traditional Mexican medicinal plant rich in phenolic compounds and pigments, yet its extraction conditions have not been systematically optimized. Using a 3² factorial response surface methodology (RSM) design, solvent type (water, ethanol, hexane) and extraction technique (stirring, decoction, sonication) were evaluated in leaves and flowers. Total phenolic content ranged from 2.77 to 97.39 mg GAE/100 g in leaves and reached up to 139.82 mg GAE/100 g in flowers, while ABTS antioxidant capacity was remarkably high in both tissues. RSM models (R² ≥ 0.70) identified water decoction as optimal for leaves and water decoction sonication for flowers, maximizing phenolic recovery and natural coloration. Metabolomic profiling putatively identified 27 metabolites including chalcones, flavonoids, stilbenes, secoiridoids, and omega phospholipids supporting the observed bioactivity. These results provide validated extraction protocols and a metabolomic baseline for muicle based functional foods, natural colorants, and nutraceuticals.

Metabolic syndrome (MetS) is a multifaceted disorder influenced by genetic and environmental factors. MetS is associated with obesity, dyslipidemia, hypertension, and hyperglycemia. Recently, attention has turned to gut microbiota, a diverse microbial community in the gastrointestinal tract implicated in metabolic diseases, including MetS. Berry cactus (Myrtillocactus geometrizans) contains polyphenols, pectins, sterols, and betalains with reported hypoglycemic, hypolipemic, anti-inflammatory, and antiproliferative properties. This study evaluated the impact of berry cactus juice concentrate (BJC) on metabolic markers, gut microbiota composition and predicted microbial functions in high-fat diet–induced MetS rat model. Metabolic markers related to MetS and comprehensive analyses of microbial 16 S rRNA gene were obtained after 140 days of treatment. HFD feeding induced a MetS-like phenotype in rats; however, BJC supplementation did not significantly reverse the main metabolic alterations under the experimental conditions evaluated. Notably, BJC treatment was associated with changes in gut microbiota composition, including alterations in dominant phyla and relevant genera such as Parabacteroides. Predicted functional analyses suggested associations with pathways involved in fatty acid metabolism and promotion of the availability of berry cactus bioactive molecules. In addition, the presence of betalains and flavonoid derivatives was identified in BJC, which may contribute to its microbiota-modulating effects. These findings suggest that BJC may act primarily as a dietary modulator of microbiota in the context of HFD-induced metabolic disturbances. Although further studies are needed to determine whether these microbial changes translate into significant metabolic benefits.
